Targeting calcium regulators as therapy for heart failure: focus on the sarcoplasmic reticulum Ca-ATPase pump

Impaired myocardial Ca 2+ cycling is a critical contributor to the development of heart failure (HF), causing changes in contractile function and structure remodeling heart. Within cardiomyocytes, regulation sarcoplasmic reticulum (SR) storage release largely dependent on handling proteins, such as SR ATPase (SERCA2a) pump. During relaxation phase cardiac cycle (diastole), SERCA2a plays role transporting cytosolic back SR, which helps restore both levels their resting state content for next contraction. However, decreased expression and/or pump activity are key features HF. As result, there growing interest developing therapeutic approaches target SERCA2a. This review provides an overview regulatory mechanisms explores potential strategies SERCA2a-targeted therapy, being investigated preclinical clinical studies.
